Multi User Wordpress Installation Issues - Subdomains not reachable - DNS error

I have a wordpress site that we would like to be able to create Multi Site Network for the sites customers. We had it working 2 days ago, unfortunately something is causing the subdomains to show a DNS error, and the site is not reachable. I'm sure this is either an error in the dns settings, the wp-config, or the htaccess file. My coder is super smart, but this one has him stuck. I'm looking for someone is HIGHLY knowledgeable with Wordpress MU sites and knows how to trouble shoot extremely well.
